Sometimes, with primers not working perfectly, you need to perform PCRs at lower temperatures. That brings the risk of primer attaching to multiple locations on the DNA and showing multiple bands after gel electrophoresis. In cases like this, you need to isolate the right DNA band. In this video, I covered how to isolate DNA bands using low-gelling temperature agarose.
