This street owes its name to the Saint-Louis-en-l'Île church located there.
The urbanization work on Île Saint-Louis began during the reign of Louis XIII, between 1614 and 1646. The island was then structured by the road currently occupied by Rue Saint-Louis-en-l'Île, then called "Grande Rue de l'Île Notre-Dame."
