Super-resolved single molecule localization microscopy data acquisition and processing tutorial
Abstract:
Single molecule localization microscopy (SMLM) methods, such as (F)PALM and (d)STORM, are valuable tools to investigate biological samples at resolutions beyond the diffraction limit. This tutorial style lecture explores the working concepts behind SMLM and will cover essential methods for processing of SMLM data, such as data acquisition, image preprocessing and point-spread function detection, localization and fitting, data postprocessing, and rendering. Identifying and removing common data artifacts will also be considered. Additionally, open-source and freely available tools will be examined with a hands-on approach using real SMLM data.
