Hematoxylin and eosin (H&E) staining is a technique used to analyze tissue sections. It involves staining cell nuclei blue-black and cytoplasm pink-red.
Steps
Place tissue sections in a staining rack
Stain with filtered Harris Hematoxylin for 10 seconds
Rinse with tap water until clear
Stain with Eosin for about 30 seconds
Rinse with tap water until clear
Dehydrate with alcohol solutions (50%, 70%, 80%, 95% x 2, 100% x 2)
Clear with xylene
Mount with a coverslip and mounting medium
How it works
Hematoxylin stains cell nuclei purple-blue
Eosin stains cytoplasm and other tissues pink-red
The staining results depend on how well the tissue was processed
