Abstract:
RPA is a rapid, isothermal method for nucleic acid amplification, but its dependence on commercial kits limits experimental flexibility. This study presents an open-source RPA system using purified proteins from the T4 bacteriophage pathway and optimised conditions to enable customizable, kit-free amplification. The platform successfully detected the SARS-CoV-2 N gene in clinical samples using lateral flow devices, offering a cost-effective and adaptable alternative to proprietary kits.
