Three neighbouring houses, streets and cars were also badly damaged in Gorse Park, Kincaidston, around 7.10pm on Monday.
A huge emergency response, including nine fire crews, raced to the scene of the suspected gas explosion and battled through the night to extinguish a blaze.
Several ambulances, including the air ambulance, were also on the scene, with medics taking two adults and two youngsters to University Hospital Crosshouse, Kilmarnock.
