Purpose
Use octanol to extract endotoxin from concentrated phage lysate. Note: If endotoxin removal is not sufficient the method can be repeated to further reduce endotoxins.
Overview
In brief, you will be adding octanol to tubes of concentrated phage, shaking for an hour, chilling and centrifuging the mixture, using a syringe+needle to pierce the bottom of a 50-mL falcon tube, collecting the aqueous phase (bottom layer) that contains your phage. In total, for about 3-4 phage samples, it will take about 2.5 hours, including 1 hour of incubation time.
*Note: octanol should be aliquoted in the fume hood, and safety goggles should be worn during needle piercing in case of octanol/phage spray.
**This video covers only the part where you remove the bottom (aqueous) layer from the tube.
