In Part 1 of our two-part series on Fatty Acid Synthesis, Professor Zach Murphy begins by breaking down the initial steps that convert acetyl-CoA into malonyl-CoA, the committed and rate-limiting step in lipogenesis. This lecture explores the role of acetyl-CoA carboxylase (ACC) and the key allosteric and hormonal mechanisms that regulate its activity.
We explain how insulin promotes fatty acid synthesis during the fed state by activating ACC and increasing citrate levels, while glucagon and AMPK inhibit the process during fasting or low-energy states. You’ll also learn how malonyl-CoA itself plays a regulatory role by inhibiting carnitine palmitoyltransferase I (CPT I)—preventing fatty acid oxidation when synthesis is active, thereby avoiding futile cycling.
