Services on this Metrolink route operate every 12 minutes with the Blue route being a full time route. The Orange route from Etihad Campus to MediaCityUK operates part time and covers this part of this route along with the Purple route to Altrincham being extended to Etihad Campus when the Orange does not operate, when combined together a 6 minute frequency is provided between Etihad Campus and Harbour City. This route uses the original Manchester City Centre route through Piccadilly Gardens interchange. When the Orange route does not operate this Blue route takes longer to travel to Eccles as it is diverted to serve and reverses at MediCityUK. Between St Peters Square and Cornbrook this service operates alongside the Purple route (Piccadilly to Altrincham), Green route (Bury to Altrincham) and Navy route (Victoria to Manchester Airport) Orange route (Piccadilly to MediaCityUK), Brown Route (Shaw & Crompton to East Didsbury and Pink route (Rochdale Town Centre to East Didsbury) and Red Route from Deansgate (Deansgate to The Trafford Centre).

The main interchange points on this route are at Piccadilly for other Metrolink services (Yellow route to Bury) along with National Rail Northern, TransPennine Express, Avanti West Coast, Transport for Wales, East Midlands Railway and Cross Country services. Deansgate Castlefield for other Metrolink and National Rail Northern services. Eccles and Ashton both provide interchange with their National Rail stations through out of station interchanges.

This Metrolink route largely uses new alignments involving reserved running or street running with little disused railway lines forming parts of the Ashton or Eccles routes. Manchester City centre is a mix of tram only rights of way and street running using alignments build specifically for Metrolink.
