The field of proteomics is gradually transitioning from cataloguing and quantitation of proteins to in-depth characterization of the protein backbone. One such characterization aspect that is been globally explored is the analyses of the Post translational modifications (PTMs) of different protein families employing high throughput mass spectrometry. The identification, assignment, characterization including structural elucidation, stoichiometry of modifications on protein back bone presents formidable challenges. Determination of such modification(s) related information provides indispensable knowledge that has major impact in fundamental research and industrial applications. Recent developments in mass spectrometry and gas phase dissociation principles provide the near complete solutions to majority of these challenges. This session will focus on the need and contribution of high resolution orbitrap mass analyzer coupled to dual and hybrid fragmentation principles for the delineation of protein/ peptide modifications. This session will also cover critical aspects involved in analyzing protein modifications such as Glycosylation, Phosphorylation, PEGylation, Disulphide bond etc. Major experimental design factors, Orbitrap MS analytical features and influence of instrumental parameters will be emphasized.
