Live demonstration of Ovum pickup and ICSI

Steps in #IVF  in Operation room and #embryology  lab: a live demo.
1.Eggs are collected under ultrasound guide. It is done under anaesthesia, so no pain. It takes 5 to 10 minutes. Lady can go home after 2 to 3 hours.
2. Aspirated follicular fluid is seen under microscope and they are separated from fluid.
3. For #ICSI the eggs are denuded( removal of cells coverings, the corona cumulus complex.
4. Husband's/partner's sperms are separated from seminal fluid.
5. Sperms are placed in a thick medium ( PvP) so that their speed of movement is reduced.
6. Good quality sperms are immobilised by hitting tails.
7. Immobilised sperms are taken or sucked by a thin glass tube( the injection pipette).
8. The denuded eggs are fixed or held under inverted microscope with the help of micro manipulator with a holding pipette with mild suction.
8. The mature eggs (MII oocytes) are in the centre of the field with Polar body at 6 o clock position.
9. Each egg is injected with single sperm with very very gentle force into the egg substance ( cytoplasm).
10. To confirm that the injection pipette is within egg, a small amount of egg's fluid ( cytoplasm) is sucked into the injection pipette.
11. Then along the cytoplasm the sperm is deposited in the centre of oocyte, and the needle/pipette is withdrawn.
12. The injected eggs are then kept for overnight and next seen for fertilization ( two pronuclei), one from female and one from male gamete.
13. They are then kept for five days to develop to #blastocyst.
14. Blastocyst are transferred then to uterus to achieve a better#pregnancy rates.
