Is it “happy”? Does it form a complex?

In this video, Stephen discusses what a “happy” sample means and how a range of biophysical techniques can be used to perform quality control on the sample. These include stability measurements via differential scanning fluorimetry (DSF) to optimise buffer conditions, measuring the polydispersity with dynamic light scattering (DLS). Structure formation can be checked via NMR or circular dichroism (CD). In the second part of the talk, Chris and Stephen explain how complex formation can be analysed with a range of biophysical techniques including analytical ultracentrifugation (AUC), size-exclusion chromatography coupled to multi-angle light scattering (SEC-MALS) and mass photometry.
The LMB Biophysics Facility houses a wide range of state-of-the-art and in-house built instruments that enable the molecular characterisation of macromolecular size, shape, mass, stability, binding affinity, dynamics and interaction kinetics.
The facility is primarily intended for hands-on research by students, postdocs, and group leaders, who prepare samples and make measurements. Biophysics supports users in advice on experimental techniques and results, training and in collaborative research.
The Biophysics Facility is headed by Stephen McLaughlin, supported by Christopher Batters.
